Gene from Pseudomonas fluorescens BlT-18 JIANG Fang-yan, HUANG Shen, LI Chun' (School of Life Science, Beijing Institute of Technology, Beijing 100081, China; 2School of Biological Science and Technology, Qiongzhou University, Sanya Hainan, 572022 China Abstract: a gene from Pseudomonas fluorescens BIT-18 encoding a protein with phospholipase B activity(Pf-PLB) was cloned and sequenced. The bioinformatics analysis results showed that Pf-PLB containing an open reading frame 1272 bp in length. The cloned gene was predicted to encode a protein of 423 amino acid residues with a theoretical molecular mass of 45.8 kDa and a deduced pl of 5.53. According to the signal peptide prediction by the SignalP, a putative signal peptide cleavage site was situated at 23 amino acids after the initiation codon. Phylogenetic and sequence analy sis revealed that pfplb is a member of a new gene family of PLBs. The homology modeling by Modeller exhibited that Pf-PLB was a B-barrel protein which consisted of 14 strands.
