Jul.2009. 25(4)?-7 神经解剖学杂志 Chinese Journal of Neuroanatomy) 被动吸烟对子鼠海马神经元内钙/钙调蛋白激酶的影响” 章诗琪陈弦何倩蒋来王清华1叶寅寅 柱丽2黄大可2贾雪梅2· ('安徽医科大学第一临床学院;2安徽医科大学基础医学院综合实验室) 摘要本文通过建立实验组孕鼠被动吸烟模型,21d后剖腹产,取子鼠脑组织,采用HE染色,尼氏染色、免疫组化染色和计算 机图像分析等方法,探讨子鼠海马神经元内钙/钙调蛋白激酶Ⅱ(CaMKⅡ)的表达变化。结果显示:CaMKⅡ免疫阳性产物分布于 子鼠大脑海马神经元胞质内,胞核呈阴性。对照组和实验组子鼠海马神经元内CMKⅡ平均光密度分别为0.310±0.057和0. 192±0.029(P<0.05)。上述结果证实:孕鼠妊娠期被动吸烟使子鼠海马神经元内CaMKⅡ表达减少,可能对子鼠学习记忆能 力有一定影响。 关键词 CaMKⅡ被动吸烟学习记忆子鼠 Effects of maternal passive smoking during pregnancy on the expression of Calcium/ Calmodulin-dependent Protein Kinase in the neonatal hippocampus Zhang Shiqi,Chen Xian',He Qian',Jiang Lai,Wang Qinghua', Ye Yinyin',Gui Li,Huang Dake,Jia Xuemei (The first clinical faculty of Anhui Medical University;2The Integrative Laboratory of Preclinie Department of Anhui Medical University) Abstract The objective of this study is to explore the effects of materal passive smoking during pregnancy on the expression of Calcium Calmodulin-dependent Protein Kinase (CaMK II in the neonatal hippocampus.Female Sprague-Dawley rats were randomly divided into the control group and the experimental group.The experimental group was subjected to passive smoking 10 days before pregnancy until ab- dominal delivery (21 days post-gestation).The fetal brain was removed,and brain tissues were processed for HE staining,Nissl staining, and immunohistochemistry.Image analysis was perfromed with the aid of computer.The results showed that the CaMK II immunoreactivi- ties were located in the cytoplasms of hippocampal neurons,and the nuclei were negative.The average optical densities of positive struc- tures were 0.3100.057 and 0.1920.029 in the control and experimental groups,respectively (P<0.05).The results confirm that passive smoking during pregnancy reduces the expression of CaMK lI in the neonatal hippocampus,which may impair the ability of leaming and memory of fetus.
